Estradiol valerate as a valerate of natural estradiol, estradiol valerate has the pharmacological effect of estrogen, can promote and regulate the normal development of female genital canal and secondary sexual characteristics, and participate in the regulation of ovarian axis function. In vitro studies with TargetValueER enhanced phosphorylation at Ser225 in MCF-7 cells showed that Estradiol(10nM) rapidly activated SphK1, a sphingosine kinase isoenzyme. Estradiol(20nM) stimulated the rapid release of sphingosine 1-phosphate (S1P) and dihydros1p from MCF-7 cells. SphK1 and estrogen receptors are the main causes of S1P and dihydro-S1P formation. Downregulation of ABCC1 or ABCG2 with siRNAs or pharmacological inhibitors reduced Estradiol(10nM) -mediated S1P and dihydros1P release from MCF-7 cells. Estradiol(10nM) inhibits the expression of miR-21 in estrogen receptor-α-mediated MCF-7 human breast cancer cells. Estradiol(10nM) activated the activity of several miR-21 target gene receptors in MCF-7 cells by inhibiting the expression of miR-21. Estradiol(10nM) increased the expression of endogenous miR-21 target genes in MCF-7 cells at the protein level, but did not affect the RNA level expression. In vivo study Estradiol(80μg/kg/day,s.c.) significantly reduced the absolute number of total peritoneal cells and macrophages in ovariectomized C57BL/6J mice, characterized by double F4/80- and CD11b- positive staining. In ovariectomized C57BL/6J mice, Estradiol(80μg/kg/day,s.c.) enhanced the expression of proinflammatory cytokines in LPs-induced TGC-induced macrophages by inhibiting PI3K activity. The pro-inflammatory effect of Estradiol is eliminated by down-regulating the activity of estrogen-receptor α in thioglycolate-induced macrophages.
