Thianaputin sodium is a tricyclic antidepressant, and its antidepressant mechanism is different from that of traditional tricyclic antidepressants. It can increase the reuptake of 5-ht in the synaptic space, and may improve the effect of 5-ht neuron conduction. No affinity for choline. The antidepressant effect was similar to that of tricyclic group, but the tolerance was better than that of tricyclic group. In animal experiments, it can increase the spontaneous activity of pyramidal cells in the hippocampus and accelerate the recovery of inhibited functions. It increases the reabsorption of serotonin by neurons in the cerebral cortex and hippocampus. Thianaputin sodium is absorbed rapidly and completely in the digestive tract. Distribution is rapid, with PPB as high as 94%. In the liver by -oxidation and n-demethylation metabolism complete clearance terminal t1/2 short, 2.5 h, very little protodrug (8%) excreted from the kidney, mainly its metabolites excreted from the urine. In patients with renal insufficiency, clearance of t1/2 was prolonged for 1h.
