In preclinical models, GIP has been shown to decrease food intake and increase energy expenditure therefore resulting in weight reductions, and when combined with GLP-1 receptor agonism, may result in greater effects on markers of metabolic dysregulation such as body weight, glucose and lipids.
Those treated with tirzepatide had an average weight loss of 15% to 20% of their starting body weight over 72 weeks (about 16 and a half months). Those taking the placebo (an injection with no medication in it) only had an average of 3% weight loss.
Type 2 Diabetes Mellitus
dosage
Indicated as an adjunct to diet and exercise to improve glycemic control in adults with type 2 diabetes mellitus
2.5 mg SC qWeek x 4 weeks initially; THEN increase to 5 mg SC qWeek
If additional glycemic control needed, increase by 2.5-mg increments after at least 4 weeks at current dose
Maximum dose: 15 mg SC qWeek
Note: The initial 2.5-mg dose is intended for treatment initiation and is not effective for glycemic control.
